I have managed to isolate different primary cell lines from osteosarcoma samples. These primary cells morphologically resemble the SaOs2 cell line but slightly larger. However, this is not enough for me and would like to confirm the identity of these cells. Literature search results are very confusing as it seems it is the lack of markers that distinguishes them from normal cells. Any suggestions much appreciated.
I have been isolating primary cells from tumour explants from patients that have managed to passage them up to 10 passages so far and still retain certain morphological characteristics. Currently looking at gene expression of markers such as osterix or survivin previously shown to be affected in osteosarcomas.However, I would like to characterise them properly using cell surface markers. The issue is, as I have mentioned in my question it is the lack of markers that distinguishes them and hence my question.
The tumour cells are derived from tumours from patients (people) as opposed to animals. The cells have been passaged for up to 10 passages so far. There is a lack of markers in primary osteosarcoma cells even in low passages, which has also been shown in the literature by pathologists and other researchers.
Hi Penelope. I have no experience with primary OS cells but with OS cell lines. There are two recent articles regarding OS surface marker which works quite well with OS cells lines.
