Probably so. But the dye is pretty exotic, its excitation and emission profiles do not fall within standard microscopy filters. Why not use a cyanine?
If you prefer Coumarin 6 for some reason, HPLC grade is probably the best that you can find. BTW do you need to load it non-covalently? It does not have any reactive group to bind it chemically.
