We are checking a protein in the urines in order to investigate its role as a marker for bladder cancer but, since its molecular weight is 45 Kda, how can I be sure that this protein come from the epithelium of the bladder or from somewhere else (passing through the glomerular barrier)? Usually the proteins present in the urine following a tubular or glomerular disease do not have this MW. It should however remember that light chain immunoglobulins can polymerize and have at that time a MW near 45

Then in order to prove that our protein come from the epithelium of the bladder we must ensure that there are no glomerular/tubular disease (we already did it). Do you think it is correct to normalize our protein versus total protein (in urines) or should we normalize it versus a specific protein in the urine (like Albumin)?

I think that, at any rate, you should normalize it versus urinary creatinine concentration, which is a good measure for the urin's concentration and easily obtainable.
Glomerular protein is albumin. You can order microslbumin to see if the protein you are getting is glomerular or not. If not and based on the MW you are including, this would be coming from epithelial cells.
