I don't have experience with the iTC200 but I assume that it behaves more or less like other ITC devices. On a TA Instuments Nano ITC for example, the sample and reference cells are kept full of distilled water if the device is not used for two weaks. If the device is not used for longer periods, it is adviced to thoroughly wash the cells, empty them and switch off the device.
In the Operating Instructions of the MicroCal iTC200 System you can find the following: "It is recommended that the power of the MicroCal iTC200 cell be turned off during extended periods of down time, such as holidays and vacations."
Additionally, the following information can be found at the following address: https://www.materials-talks.com/how-to-shut-down-microcal-instruments-for-extended-storage/
ITC200 Manual
After the last run, perform a cell and syringe wash. Then fill the sample and reference cells with distilled or deionized water. You can also use a 0.02% sodium azide solution to fight bacterial growth in the cells.
After this:
Close the MicroCal ITC200 control software.
Switch off the power at the back of the ITC cell unit.
Power off the controller computer.
Clean and rinse the cell filling syringe and leave it to dry with the plunger removed from the syringe.
If you are not going to be using the iTC200 for a few weeks, yes make sure the sample cell and reference cell are completely cleaned (use the recommended detergent 14% Decon 90, soak at 60 deg C for at least 30 minutes, cool cell and rinse with many ml of water), empty cells and power down the instrument and computer. Also make sure the injection syringe and the syringe you use to fill the iTC cell are cleaned and dry. You should have the cells covered, or you can have the injection syringe placed in the cell to prevent particle sin the air from getting into the ITC cells.
Thank you very much Luminita Duma and Verna Frasca !
Verna Frasca : so you suggest that I perform the detergent soak and rinse before I switch off the devise for a longer period, correct? Concerning the syringe- can I place it in the washing station? Thank you again, ladies!
Elena Krachmarova yes do the detergent soak and all the water rinses before you turn off the instrument. If the ITC cells are dry, but still contaminated with protein, then they will be more difficult to get clean. You can keep the injection syringe in the washing station, in that case be sure the ITC cells are covered. The instrument came with a cap for the cell, if you no longer have it place a small beaker or something else over the cells.
Thank you very much, Verna Frasca ! I'll do as you recommend. I still have the cap for the cell, and I always use it to cover the cell. Thank you, again, for your time! Best regards, Elena
