Both semi dry and wet transfer will accomplish the same thing. I prefer semi dry myself, but others prefer wet. Either method will work.

Thanks for the reply. But I heard protein which more than 100kDa weight should prefer the wet transfer and less than 100kDa go for the semi-dry. does it right ?

We only use wet transfer - and we get decent transfer of the whole Molecular weight ladder. With either system, you may need to optimize the protocol for your particular target(s).
