In the differentiation of pluripotent stem cells to hepatocyte-like cells, some protocols include hydrocortisone as a reagent, but some others use hydrocortisone 21-hemisuccinate sodium salt. It seems that they both inhibit the immune response, but are there some difference between these two molecules in differentiation and maintenance of hepatocytes?
You have to carry out the expts.separately and observe the responses and learn
the salt solves much easier in medium while the non-salt is difficult to solve. Maybe use DMSO
I believe the hydrocortisone 21-hemisuccinate sodium salt is used mainly for its solubility in water. Hydrocortisone must be put in different solvent such as DMSO before adding to media. I have used both compounds in hepatocyte experiments, and have not seen a difference.

If you wish to replace human hepatocytes by the closely related HepaRG cell line, solubility will not be a problem anymore as it differentiates in hepatocytes in the presence of DMSO in the cell culture medium.

I only used hydrocortisone 21-hemisuccinate sodium salt. It is easily dissovled in water.
