I want to perform immunophenotyping to check for a number of cell markers in the spleens of vaccinated mice. To check for memory B cells, I see that there are a number of cell markers. Because of the cost of the antibodies, I cannot afford to check for all the markers for memory B cells. If I check for the B220 (CD45R) marker alone, will that be sufficient to prove the presence of memory B cells?
Yes, in addition, memory B lymphocytes include CD45RO+CD62L+CCR7+ T cells (central memory T cells, TCM) and CD45RO+CD62L-CCR7-T cells (effectormemory T cells, TEM).
Since you want the sub-population of memory B cells, it might be more important to also include the memory marker CD27.
Downregulations of CD45R (B220) marker basically indicates the cell differentiation into plasma cells and memory cells. In general, CD20, CD27, CD40, CD80 and PDL-2 are good markers if you are interested to define various subsets among memory B cell population. You may also include IgG, IgA and IgE if you are interested in cell surface immunoglobulins.
