I would like to label monocytes with a marker visible with an MRI or PET. What is the best source and/or easiest for the cells? Should I just go with primary cells (e.g. from the peritoneal cavity or spleen) or is there a cell line that would work as well? I will be working with a C57BL6 mouse.
I would use bone marrow-derived DCs. Using I10+GM-CSF. To get more you can pre-treat the mice with Flt-3 ligand.
For mononuclear cells you could do a ficoll/percoll gradient of blood. You could also isolate them from spleen or bone marrow depending on what kind of mononuclear cells you are looking at.

Hi, I would use monocytes from the bone marrow, positively sorted using biotinylated anti-CD115 Ab and anti-SA MACS beads. Usually, 5x106 monocytes can be obtained from a single mouse.
