I would start with screening what signaling pathway/s will alter during the transformation from what you call "normal cancer cell" to a "resistant cancer cell" upon treating with Vemurafenib. I know this sound crazy when it comes to analyze multiple pathways but you can do an IPAD assay (immuno paired antibody detection) which will cover about 20 signaling pathways including many anti apoptotic pathways and Braf/MEK/ERK signaling which is the target for Vfenib. Or you can do ELISA. From these experiments you should be able to identify the proteins alter during the process of gaining drug resistance. Then you can try to overexpress a target that you will identify in above experiments. I guess you can have multiple options from this.
