I've isolated exosomes from a neuronal cell line and I've isolated exosomal RNA and I intend to perform qPCR. So, I was wondering which mRNA could be useful to attest the purity of my exosomal sample, to exclude the occurrence of cellular contamination? Is there any mRNA that is only present in exosomes but not in cells?
While some mRNAs may be relatively enriched in exosomes or other small extracellular vesicles, extracellular transcripts originate in the parent cell and should thus be detected in both exRNA and cellular RNA. A useful marker would have to be highly enriched, indeed, and I for one am not aware of such a message...which isn't to say it doesn't exist, of course! Still, it's difficult to envision a sequestration of a particular message into exosomes that's so rapid and complete that no copies could be detected in the parent cell.
