- Incubated in anaerobic condition for 24-48 hrs for growth. (Done)
After the growth, we are intended to take single colony in TSB,
-> then to shaking incubator.
-> After that, glycerol stocking.
But do the anaerobes single colony would survive, after taken to shaking incubator?
Would they survive in such aerobic condition?
It depends upon whether your organism is a true anaerobe, in which case it would not grow with aerobic conditions, or a facultative anaerobe which will grow under both aerobic and anaerobic conditions and hence should grow in your conditions.
To your question What will happen to anaerobic bacteria, if taken to shaking incubators? : It depends on conditions during shaking. If the conditions will be strictly anaerobic it happens nothing.

I think it depends on the material that you are using.. Quantity of glycerol need for sticking may differ
