Hi all, I just want to know that, if we keep the siRNA in a -20 degree freezer (as mentioned in the product detail), what will be the average life of the siRNA? Can we store for more than one year, if not opened?
Gautam Kumar this partly depends on what you are storing your siRNA in. A six month shelf-life is about the average. I would use -80oC if you are planning anything longer.
Form my personal experience, I would expect a small amount of degradation after about a year, but make sure the buffer is actually frozen solid, not remaining liquid. I would suggest verifying the condition of the siRNA by HPLC.
I think electrophoresis should work for that, but I personally prefer HPLC because, in my experience, it’s easier/more accurate for quantifying, at least with the instrumentation we have.
siRNA should not undergo more than 5 freeze-thaw cycles, and can usually be kept stable for more than 6 months. If degradation needs to be considered, the integrity of the siRNA duplex can be evaluated on the analytical PAGE gel.
