Basically I want to see the macrophage cells I have cultured on these opaque materials under a microscope, so what kind can I use? I tried viewing them under a Scanning Electron Microscope, but I have noticed that my cells were damaged due to the process of fixation. Can someone help me find the best microscope that will help me solve my problem?
Why not try to improve your fixation and processing procedure and stick with SEM? What leads you to the conclusion that the cells were damaged? This should be a fairly straightforward procedure.
When i compare SEM images of my control and my sample, they are completely different. It seems more like cell debris and scattered parts of cells on my material.
Have you ruled out the possibility that your cells are not surviving on your material? Perhaps it's too rough?
