I need to quantify genes expression for which i need to perform qPCR but the problem I am facing is difficulty in RNA extraction from feed and soil samples. Can I use genomic DNA to get some level of expression using RT-PCR ?
if I get you right, you want to determine/estimate RNA-expression levels based on genomic DNA e.g. copy numbers of your gene.
If I am not mistaken, this is not 100% possible, because of way too many possible influences you have from DNA to RNA . It is nearly the same thing as if you would like to predict protein level based on RNA or DNA, you may get a hint on it, but you never can be sure.
among other things, there are 3 important partners in cells, DNA, RNA and proteins. DNA will give by transcription RNA that will give by traduction to proteins. between each steps, there are a lot of regulation points resulting in the fact that all genes are not equally expressed in RNA and at the end in proteins.
one copy of gene A could give you 10 transcripts whereas one copy of the gene B could give you 100 transcripts...
it means that you can't have expression profiles from only DNA data.
if you succeeded to extract DNA, even though the protocol is slightly different, there is no way you can't extract RNA.
