I am intending to perform antibiotic susceptibility assay on Staphylococcus aureus cultures, by broth microdilution method. Now usually Resazurin Sodium salt (Alamar Blue) is used as a marker of cellular viability in these types of assays. I want to know that what can be the most suitable alternative of Resazurin Sodium in this case, as we are having unavailability of Resazurin in our lab.
For bacterial viability I would also recommend visual analysis in 96 well plate or even plating (of a dilution) and counting the colonies on the agarplate (CFU) to define the minimal inhibitory concentration.
I usually use 96 well plate and culturing for MIC and MBC, I never hear about Alamar Blue.
Visual method work well, resazurin would get metabolized very soon by S. aureus and you have to take fluorescence as soon as possible. Most of labs worldwide perform this by visual method. I agree with above opinion.

The best accurate method for obtaining definitive results is culturing from all wells of the 96 well plate to can easily detect your MIC and MBC values.
