I am performing antibiotic susceptibility study against S. aureus by broth micro-dilution method using 96 well plates and using MTT dye as a marker of viability. Usually after dispensing MTT dye in each well and going for an incubation of 3-4 hours I usually take OD for each well of the plate on microtiter plate reader in order to quantitatively determine the viability of organisms. Is there any formula by which I can calculate the percent inhibition of organisms in each well by utilizing the values of OD?
To determine the percent inhibition of bacterial growth use the formula: [(Ac–At)/Ac] × 100
where Ac is an average of six replicates of light absorption values at particular wavelength (commonly 510 nm) of the negative controls, and At is an average of six replicates of light absorption values at particular wavelength (commonly 510 nm) of the samples.
