We have 24 amino acid in our peptide sequence.

24 amino acid- (24×110) = 2640g/mol =Molecular weight.

Molecular weight of out aptamer- 24,009.5

We are taking 6.46 µl of protein from stock. (116 µg/ µl)

Then amount of protein- (116×6.46) = 749.36 µg = 749.36 × 10-6 g

Molarity= 749.36 × 10-6/ 2640

= 0.283 × 10-6 mol

= 0.283 µM

We have diluted it with binding buffer so, (50/6.46) = 7.73

Now, (0.283/7.73) = 0.0366 µM

We are taking 10 µM aptamer final concentration.

So, molar ratio will be- (10/0.0366) = 273.22

Currently we are using molar ratio aptamer to protein = 273.22 : 1

Whether this ratio correct?
