The raw cells were grown in Dulbecco’s Modified Eagle’s Medium (DMEM, high glucose, Glutamax supplement, pyruvate Gibco) supplemented with 10% fetal bovine serum (FBS), 2 mM of L-Glutamine (Gibco) and 600 μL 50 mg/ml Gentamycine (Gibco) (total of 30 mg)
--> what kind of polarization ratio would the raw cells have in this media.
I was also wondering what the effect of antibiotics could be on the raw cells (except the cryptic contaminations). Because 3t3 media seemed to shift the polarization of the cells into the M1 subtype.
3t3 media:
The 3T3 fibroblasts were cultured in Dulbecco’s modified Eagle’s medium (DMEM, GlutaMAXTM)
supplemented with 10% fetal bovine serum (FBS,heat inactivated), 4,5 g/L glucose (Biowhittaker), 2 mM L-glutamine (Gibco), 100U/ml pencillin (Sigma) and 100μg/ml streptomycin(Sigma).
The only difference between the two media is the use of the antibiotic...
