Concentarion of acridine orange 6 mg/ml is sufficient to fluorescent microscopy, Acridine Orange solution is stable for several weeks when stored at 4°C and in the dark.
I used 3µg/ml for zebrafish embryos for 15min at 27C, then 3 washes of 10 minutes each. Whether you are imaging cells or embryos, imaging has to be fast as photoactivation of AO by UV light will trigger some amazing explosions of the lysosomes! Nice if this is what you want to see, but not so nice when it happens before the imaging is done...
