I am working with cancer cells and assess the effect of different compounds on NFkB pathway and apoptosis. As I observed phosphorylation of p65 upon treatment and a rescue of apoptosis by adding 2 different NFkB inhibitors, I assumed an unexpected involvement of NFkB as inducer of apoptosis. Not in line with this, I see a decreased activation of NFkB activitiy by using a commercially available NFkB reporter assay kit (Promega). Does anyone of you have a possible explanation for this? I would apprecciate any recommodations.
