As we are facing difficulties with transfer of MSC to chamberslides right now we dont have a usable method for staining of cultured MSC with the conventional fluorescence microscope. Do you have any recommendations?
If it is just MSCs, you can stain with any common stain. Hoescht works well for live cells, gives a nice picture of the nuclei. You can also use a membrane dye such as DiI or DiO before transferring or a cytoplasm dye such as calcein AM. If you want specific surface markers, CD90 is typically expressed at pretty high levels and can be stained live or fixed.
Thank you James! Can you recommend a way to do the staining with a conventional fluorescent microscope (not inversion). Chamberslides for some reason do not work, the cells do not attach. Ibidi-chambers are not practical with our microscope.
