For obtaining these compounds from plant extracts u should use aqueous solvents like 70% ETHANOL, 70% METHANOL OR 70% ACETONE or you can also use hot water (60-80 degree C) for obtaining compounds of interest. Then using HPLC or TLC you can check the yield of desired compounds in the extracts. Thereafter yoe should proceed with fractionation or partitioning by using solvents of differnt polarity.

this can be done by column chromatography eluting with low polar solvent ot higher. usually sterols will be obtained in low polar solvent fractions. some sterols (like sigmasterol and sitosterol) will be present in 70:30 pet ether: ethyl acetate fraction. flavonids can be obtained in high polar solvent fraction.
From Crude extract , you can make a liquid-liquid partitioning ( LLE ) . starting from a quantity of Crude extract diluted in a solution of 15 % ethanol. This partitioning solution with 3 x 200 ml of hexane to remove polar compounds , then extracted with ethyl acetate, concentrated where the phytosterols and some non-glycosylated flavonoid . Will be concentrated in the aqueous fraction oS Well Polar compounds , eg flavonoids glycosides . The Next Step to obtain these compounds would be A chromatographic separation , which CAN be through RP- HPLC with gradient methanol : water , this kind of separation you do not need to dry the solvent fractions , OR the other hand , by column chromatography with silica , however , you need this Secaras fractions obtained by LLE ( Fraction acetate with rotary evaporator ) and iN aqueous fraction lyophilization . and the gradient of this separation can be scaled starting from hexane : ethyl acetate : ethanol .
