Dear folks

I am facing a problem with my HUh7 cells grown in DMEM (high glucose, L-glutamine +, sodium pyruvate -) with 10% FBS , Non-essential amino acids (1X), L-Glutamine (2mM), Antibiotic.

After the 3 or more passages cells start loosing their morphology and shedding lots of debris. i am hereby attaching pictures of healthy v/s affected cells. Kindly note my incubator maintains following parameters:

CO2 - 5%

Humidity 70-80%

temperature 37C +/- 1

What can be the problem of such a drastic cell death as their is no visible biological contamination in the cells except lots of debris after few passages?
