I am specifically interested in synthesizing cyclotides with a small peptide graft as described in the experimental procedures of the paper: (Rasmus Eliasen et al. "Design, Synthesis, Structural and Functional Characterization of Novel Melanocortin Agonists Based on the Cyclotide Kalata-B1"). SPPS is the de facto methodology for production of these proteins. I have a $1000 budget for this procedure. Does this process require a high amount of laboratory training and/or cost compared to E. Coli expression of a more standard protein type?
