There are some studies on fused drug molecules. Sultamicillin is one of the example for this type of prodrugs. The drug would be hydrolized and reveals two different active molecules which is sulbactam and sulfapyridine. What is the limitations and requirements for the duration of producing the fused prodrugs?
The method of attachment of the two drugs should be such that the mechanism that allows the two drugs to be separated (esterase in the case of sultamicillin) can perform its function on the necessary time scale.
Since the 2 drugs would be released in a 1:1 stoichiometry, it would be necessary for both of them to have the necessary potency at the concentration achieved.
Both of them would have to have appropriate pharmacokinetics and pharmacodynamics when released in the same amount to achieve the necessary efficacy.
The drugs should not be antagonistic in their action (against bacteria in this case).
In the antibiotic field, there are many 2-drug combinations, such as a beta-lactam and a beta-lactamase inhibitor (like sultamicillin), but they are usually administered as 2 separate compounds. This allows the amounts of the two drugs administered to be different if that is required.
