I used indomethacin as control drug in cotton pellet induced granuloma model using rats. Currently, I used 5 mg/kg of indomethacin. I prepared indomethacin in 10% DMSO. However, all rats died at day 3 (the duration of treatment is 7 days). From the observation, the rats refused to eat and the stomach become blotting. After post-mortem, I found that the lining of stomach become thin. May I know the right method to give indomethacin to the rats as most of studies using indomethacin as control. For your information, I fasted the rats before started the experiment. On day 1, after cotton pellet was implanted, the rat was given indomethacin orally. Then, the food was given.
