I am trying to isolate an organism from the coal sample and did some subculturing methods to let the organism develop more. how to identify the unknown bacteria in the culture plate using 16s rRNA technique???
A simple 16s rRNA sequencing is not enough for reliable identification of unknown bacteria. Before that, you have to do some preliminary tests to guess what group or family your bacteria is belong? Different bacterial groups, need different DNA extraction methods, and after that different sub-sequencing (other parts of genome) is needed for correct identification. Without early detection, with raw 16srRNA sequence you can not find correct answer in BLAST or phylogenetic tree.
If u have single colony you can go directly for colony PCR and DNA sequencing.. It is the most reliable technique for the Identification of genus and species.. But it is little expensive...
