I was wondering if anyone could tell me about the stability time limit for dU-ssDNA at -20C. I am currently developing a synthetic phage display library, and I have recently extracted uracil-containing single stranded DNA (dU-ssDNA) from the M13 bacteriophage that I am using. The DNA contains uracil because I am growing up the bacteriophage in CJ236 E. coli, which is dut- and ung-. Deletion of these two genes facilitates the incorporation of uracil into DNA. Moving on, I have had a few aliquots of dU-ssDNA frozen away at -20C for about two months. Instead of starting from scratch, is it possible that this dU-ssDNA is still stable enough to use for Kunkel Mutagenesis with a degenerative oligonucleotide? If anything about my question or experiment setup in general is unclear please say so, and I will do my best to clarify. Thank you in advance for any and all help.
