Although, triple sugar test (mannitol, sucralose and lactulose) has already been employed for estimating colonic permeability in animal and human but it requires the use of Gas chromatography that is quite expensive technique. I need a method which is less expensive and doesnot require any surgery on animals. Any lead would be appreciated

we assessed the in vivo intestinal permeability using fluorescein isothiocyanate (FITC)-dextran. You can give your mice DX-4000-FITC (FD4000) by oral gavage (500 mg/kg body weight). You collect blood samples after 3 or 4 hours. Then, you quantify DX-4000-FITC concentrations with a fluorescence spectrophotometer. You can collect blood from mice at different time points, without any animal sacrifice.

In addition to fluorescein isothiocyanate (FITC)-dextran method, you can use the magnetic resonance imaging (MRI) for the assessment of the intestinal permeability based on the oral administration of clinically approved gadolinium (Gd)-based MRI contrast agents.
