I am using a Brendel assembly of UA6 detector and Fractionator. As we dint have an engineer assemble it for us, we tried our hands at it. I wanted to know if there is a set program for obtaining fractions from the fractionator? Would anyone mind sharing it?

It really depends on the flow rate. In my experience, when breaking down a sucrose fraction for polysome analysis, I use a flow rate (i believe 0.5 ml/minute; if my memory serves correctly) that allows for nice resolution of absorbance peaks. Moreover, I adjust the fraction volumes to obtain 12-18 fractions that span the entire sucrose gradient. Please let me know if you have further questions.

We use BioComp fractionator for the same purpose. We use 0.3mm speed, 3mm distance for SW55 tube. 14 sec of fraction collection gives around 300- 400 ul fractions. But depending on the tube eg SW41, you can vary the fraction volume.
