Hi all, I recently purchased Ni-NTA Magnetic Agarose beads (from MedChemExpress, HY-K0241) and would like to immobilize a His-tagged protein. After immobilization, I plan to perform an ELISA to verify successful binding, followed by further applications involving oligonucleotides that interact with the protein.

To ensure the best performance and avoid extensive optimization, I would really appreciate any information shared regarding:

• Suggested protein concentration for immobilization

• Any relevant protocols or tips for ELISA setup with these beads
