Hi,

ACK lysis buffer is great for non-nucleated RBCs, but not good for nucleated ones.

Birds, reptiles and fish have nucleated RBC so the traditional ACK doesn't work well (nor does water alone or BDFacsLyse).

I found a recipe on ResearchGate making it with MgCl2 and sucrose etc. I was wondering if anyone could confirm the recipe and tell me how it is used (time, temperature, diluent post lysis stage).
