For mAb purification process we performed UF/DF as a final step. Firstly we used small scale cassette (88 cm2 ) as a trial and we did not observe any recovery decrease. Then we pass to 0,1 m2 cassette and repeated experiment with same parameters. During ultrafiltration and diafiltration we took samples to check concentration and recovery. When the results came, we saw that there was no loss in the sample taken after ultrafiltration, but protein was lost in the sample taken after diafiltration. Additionally we did not observe any pressure increasement and permeate flow drop during diafiltration. As a calculation nearly we lost 40% of total protein after diafiltration. We also checked the permeate samples (UF and DF) to see is there any protein and we did not observe any protein. We did not expect such a large loss of protein as we did not encounter a problem in our small scale experiment. Although no flow rate decrease or pressure increase was observed, could such a high amount of protein bind to the cassette or could there be another reason for the loss?
