Most of the researches I went through suggest increased crystallite size after being loaded with a drug. However in a unique case, I have observed reduced average diameter after drug loading. Can there be a justifiable reason behind this? Kindly enlighten me on this topic.
What type of NPs are you using?
What is the drug you used?
What is the method you used for determining the NPs size?
Please answer these questions which can help answering your question.
1. have you washed your product properly?
2. The free-floating/stereo-oriented surface groups adopt to the confined area of the encapsulating/solid vesicle.
3. The detachments of weakly (physical forces) bound entities on the surface.
4. Shrinkage may also be due to electron-cloud density/orbital changes!
5. For average diameter, low levels of internal de-aggregation due to physicao-chemical parametric changes.
Mohammad Kooti , Various types of metallic NPs were synthesized including Ag, Au. Drug used was 5-Fluorouracil and the method utilized for size determination was XRD based Sherrer equation.
Riaz A. Khan , yes, synthesized NPs were washed with ethanol prior to drying in hot air oven. The reasons mentioned by you seem very logical. Can you kindly provide some relevant literature that could support these?
The reason can be attributed to the drug which may act as surfactant and prevents aggregation of Ag NPs. Specially when the loaded drug is high.
In this case, one expect that after releasing of the drug from Ag NPs their size will increase.
May be you used a capping agent when you synthesized the metal nanoparticles before binding to drug, which gave them a certain size you measured it first . And later, when you binded them to the drug they released from the first capping agent and the drug itself acted as a new capping agent for the metal nanoparticles. In this case may be the new drug capped particles is smaller than the first synthesized particles due to the difference between their molecular weights. So when measured again it gave smaller size.

To prove the reason I have mentioned you don't have to refer to another research. You can prove it yourself. Just let the loaded drug release completely from the surface of the Ag NPs and after a while filter the NPs and finally take an XRD from the sample to see if the size of NPs grow.
