we use an automatic infiltrator in our lab which can be programmed to change alcohols at set times and can let the tissue sit in wax for a set time. does anyone know the optimum times for alcohol washes and wax incubation?
i've done liver samples before, for which i've used 1 hour each in 100%-30% alcohols, 1 hr each in citrosolv (twice), and half an hour each in wax (twice).
i want to know if there is a different timing system for kidneys.

There should not be much difference between liver and kidney as far as the samples have the same or similar size.
I would recommend to prolong paraffin-infiltration also for at least one hour each.
What are your ethanol-steps in detail. Here we start with 50%-70%-96-96-100-100.
If the biopsies are thick I would recommend extending the steps of embedding. In order to a proper paraffin-infiltration we used ethanol 70% 12h, 96% 12h, 100% 12h, xylen or other similar organic solvent 3x30 minutes, paraffin 1 - 3h, paraffin 2 - overnight and paraffin 3 - 3 h. If the biopsies are not thick, the procedure of paraffin embedding is well documented.
I am using parafin blocks to study kidneys and just after dissection I place the organs in 10% formalin overnight and the next day I begin the ethanol steps 75% overnight-80%1h-95%1h-100%1h-xylene1h-1st Paraffin 1h-2nd Paraffin overnight)
As long as Ganesh works with an infiltration-instrument, the times with 1-2 hours will be sufficient.
