I am using angiotensin II to stimulate rat VSMC proliferation. The procedure is as follows: Seeded the cells in the plate with growth medium; Incubated for 4h; Removed the medium; Added the incomplete medium and did the stimulation with two different dosage Ang II (100nM, 1000nM) for 24h; Tested the proliferation by BrDU. However, there was no significant change in the cell proliferation. I am wondering if there might be some problems in the stimulation procedure. Thank you!
