When any compound is newly synthesized and characterized by various techniques. we should declare the assay of that compound. I just want to know what is the procedure to be followed to declare the assay if you doesn't have any reference standard which is normally required to declare the assay of the compound
You can test that compound in animal model for finding potency and use established cell lines to cross check the cellular effects, if it shows some activity against any type of morbidity or abnormality than it will be a much potent compound. Potency means over all efficacy of compound that can be defined to ensure disease curing capacity or to sub side any physiological, biochemical and any type of abnormality if body systems.

thanks for your guidance.My problem is related to active content of a compound only. I mean it is related to assay of a chemical compounds.
It depends on your compound and the definition of "potency". Employing a primary method you do not need a standard.
