I am trying to determine the Minimum Inhibitory concentration (MIC) for Several Antibiotics against 13 different Antibiotic Strains. I came to know that this can be determined through the 96 well Microplate conveniently and efficiently. However, Majority of the literature is reporting Agar plate diffusion method for the same. I want to confirm what method should I use.
I think MIC in 96 well plate is the more accurate and acceptable method.

It's a very general confusion about the selection of techniques. It depends how you would like to express your results. Disc diffusion is generally qualitative approach and much cheaper than antibiotic dilution technique which is more quantitative.
CLSI favours both the techniques. But if you wish to know the exact value of MIC go for dilution method. It can be helful further by avoiding antibiotics abuse if you know the MIC exactly for a particular strain against a particular antibiotic.
I think the disc diffusion test is suitable to check if the bacteria are susceptible, intermediate resistant or resistant to your antibiotics. But for the MIC determination I would use the microplate dilution method because the mixture of the antibiotics is more accurate with a liquid medium than with an agar medium in a petridish.

I also think micro plate dilution technique is better for MIC determination.
In case of unnkown antibiotic break points the two methods should be used and compared to set the lilits of inhibition. For usual known detrmination of MIC, the microplate dilution method is the most rapid and suitable for MIC tests.

I would say that 96-well plate is better, easier and faster to get the results. However if you get a contamination, it would be difficult to notice using a 96-well plate.

How results should be expresses if MIC is done in 96 well plate?. say, I got the MIC value of 5mcg/0.1ml of double diluted broth in 96 tray. Should I multiply it by 10 in order to convert to 1ml?
