We want to measure Raman spectra of tissue samples. However, we can not take fresh samples. We have to store them first. What is the best practice? Snap freeze? Snap freeze with optimal cutting temperature compound? Paraffin embedding?
We are in between first two. What do you think?
Ugur, in my experience with Raman spectral analysis and porcine livers, I documented in the attached literature steps to take to clean out blood remnants, treat with anticoagulant and store on ice during transplant and prior to measurements. In the literature review of this same manuscript, a little more insight is given on procedures that are in place pending organ transplant.
Per your question, snap freeze will be idea for measurements done within 8-12 hours or treatment of samples for long term storage in a -80 C.
