This is my particular survey to know how long people usually store total RNA samples at -80°C prior to library preparation for RNA-seq analysis. I would like to know if anyone has experience with long-term storage and had any problems with this kind of analysis.
It doesn't matter how long you are storing. Because the stability may vary depends on the storage solution and the way you have isolated RNA.
There is a thump rule before preparing library is to check your RNA quality always If you ok with the quality then go ahead.
You can also check the quality come stability test (which i personally do). After RNA isolation, aliquat about 5-10ul of RNA and keep it at 37 C for 1 hr.
Then compare its quality with the stock. If there is any quality change, there is some nuclease presence. In that case its better to process the sample as soon as possible. If there is no degradation after 37C / 1hr, you can store it 1 week - a month with DEPC water or NFW

Well-isolated RNA is quite stable in -80 degress (RNAse free water). After 3 year of storage in these conditions I found only minor drop in RNA integrity. Besides RNA integrity, I do not know of any other significant potential confounding factor, that could influence sequencing. Just check RNA concentration right before sample preparation. There could be some small changes for example due to binding of nucleid acids to tube walls.
