I am using a Cre-ERT mouse model and giving mice tamoxifen by oral gavage at 8 weeks of age. 1 dose of 6 mg of tamoxifen does not generate enough Cre activity. Does anyone have any recommendations for a greater dose?

First, you can check in the litterature if someone else has already used the same mouse model and see how much tamoxifen they used to induce the recombination.
I don't think a single dose of tamoxifen will be enough. It's even sometimes used to induce partial recombination. So instead of increasing the dose, I would recommend to repeat the gavage for several days.
For my Cre-ERT2 model, I do 5 days of gavage with 150mg/kg of tamoxifen (e.g. 4.5mg for a 30g mouse), once per day, . The treatment is done in 8 weeks-old mice.

Our study mice received a daily dose of 2.5mg of tamoxifen (Simga-Aldrich) diluted in a mixture of 10% ethanol (VWR) 90% sunflower oil (Sigma-Aldrich) by intraperitoneal (IP) administration, for 10 consecutive days (25mg total).
