I would like to use CCCP as a positive control for JC-1 experiment on colorectal cancer cells. What concentration of CCCP should I use at the beginning and how long the cells should be treated?
Practical use of at 0.05 mM is recommended; i.e., 10 mM of FCCP is dissolved in DMSO, and 1/200-volume is added to the medium (final DMSO conc is 0.5% (v/v)) (see file). It is noteworthy that only FCCP is effective in mammalian-cultured Caco-2 cells (see file).
You should do a dose response assay . Usually in the range of 0.1-20 micrograms /ml...but do not use DMSO to prepare cisplatin
