I wanted to detect changes in MHC Class 1 expression using qPCR in MCF7, but MHC Class 1 is categorized as classical (HLA-A, B, C) and non-classical (HLA-E,F and G) and some of it are highly polymorphic (especially HLA-A). According to immunohistochemistry techniques involving mAb and pAb usage to stain tissue samples or solid tumour mass, the results varies in terms of several phenotypic expression (loss of HLA-A, combination loss of HLA-A and B, increase in HLA-E etc with respective to their studies such as MHC Class I in metastatic or increasing tumour mass). So, is there any single target that is definite to portrays expression changes of MHC-I (via qPCR) in MCF7 or MDA-MB-231 either via HLA or other molecules (eg; Beta2 micro-globulin - but probably unsuitable due to its mutation mostly in post-translational in cancer cells)? Or will I have to target more than one HLA type for conclusive expression changes in qPCR studies?
