I am running an ELISA to check for cytokines but the difficulty I am facing is I got some reagents without standards. Is it possible to run interleukin 4 (IL-4) or interleukin 10 (IL10) ELISA without standards? If it is possible, can anyone be kind to share with me his/her experience. Can I get a protocol or any material or link that could be of help to me. Many thanks.
It's not possible since you have to make a calibration curve every time you run cytokines by ELISA. It's not correct to use another calibration curve since they don't reproduce your very exact conditions.
For a relative comparison (inhouse needs), you could run an ELISA without standards. But to get an exact concentration you MUST have the standard curve run along with the samples.

you can run ELISA without STD but it will be a qualitative method, therefore you can contact the company or any immunological institute to provide you with STD.
Without standard data will be qualitative, but for absolute quantification, you need standard, at least, one known the quantity of IL-4 or IL-10 PROTEIN (recombinant protein of the same organism). Then by the unitary method, you may deduce a rough estimate. But it is indispensable to run a range of known conc. for preparing std curve.
Why don't you try some RT-PCR studies first so it will give you some evidence to buy the reagents that you need. Otherwise you don't have to carry on if you don't see anything by PCR.
