There are still many well developed methods and ready-to-use kits for isolation of viable and functional mitochondria. However, it is difficult to choose the best way to achieve high quality mitochondrial yield. The isolation of functional mitochondria from human stem cells is a part from one of our projects. I was wondering if somebody that has an experience with isolation of mitochondria by differential centrifugation and differential filtration would give an advice. What of these approaches is better in this particular case ?
