I am about to begin a project where I will be generating organoids from prostate tissue. I have read several protocols that use growth factor reduced Matrigel. I was speaking to a rep about it and she asked what was the minimum percentage I required. I had assumed that it was just test a few batches to find one that works, but if there is a way of eliminating batches that certainly won't work (ie too low a percentage) then that would be a great start. So, what do you ask for when ordering/testing a fresh batch of Matrigel to grow prostate organoids?

Also, can someone suggest how to test batches? Obviously you can't use patient samples because you don't know if they will grow. Will growing cell lines as organoids a sufficient test? Or would growing mouse prostate organoids be better? And again, is this a good test or are they less fussy?

Lots of questions, but I'd appreciate any help and advice anyone can give me.
