I want to place a (10 uL) drop containing a known concentration of conidia of the fungus C. rosea in a malt extract agar petri dish (⌀ = 9 cm) to investigate mycelial growth under the influence of antifungal compounds. The Petri dish will be incubated at 25 °C and the antifungal effects will be monitored by measuring the diameter of the mycelium. However, I would like to perform this within a week. What would be a safe concentration of conidia for this study? In advance, thank you!
It depends on many circumstances like the type of medium, temperature, pH etc...
The test you described is not the best to evaluate the effects of a poison on mycelial growth. The poison in the medium will affect both conidial germination and radial growth of the colony. It will be better to collect a disk of colonized medium from the margin of an actively growing colony of the test fungus and use it instead of the conidial suspension drop
As mentioned by Prof. Giovanni, it is very important to use an actively growing colony, and then you can get a nice mycellium growth with a minimum spores number.
