I am using ex229 (compound 991), (10 µM), as +ve control for AMPK activation (phosphorylation) in J774 cells (macrophages), but it is not working (Western Blot).

It is showing +ve effect on p-ACC but not on p-AMPK.

My test compounds (natural products) are showing +ve effect on both AMPK & ACC phosphorylation.

What can be the reason? Can I increase the concentration of 991, more than 10 µM?
