Aprotinin is a protease inhibitor and I have my sample in very dilute form. I can concentrate and use but need to know what is the least amount of aprotinin this assay can predict.
The answer would depend on the conditions of the assay (buffer composition, pH, salt concentration, temperature, the signal-to-noise ratio in the assay, and most importantly, the protease being inhibited. If, for example, the concentration of aprotinin needed to produce 50% inhibition in the assay is 20 nM and your assay allows you to detect 50% inhibition convincingly, then 20 nM aprotinin would be detectable.
