I am looking to see if anyone has labeled a surface protein and simultaneously confirmed that the cell's plasma membrane is intact and not permeable by probing with a specific antibody used to detect intracellular antigens. It would be great if you could also give me a reference or mention a gold standard method to follow.
P.S. I am not looking for membrane-specific or lipophilic dyes to directly target the membrane or any other non-protein labeling dyes.

Sytox Green is a membrane-impermeable dye, whose fluorescence is enhanced upon binding to DNA and other nucleic acids. It is frequently used to test for membrane integrity in cells. It will typically stain dead or dying cells.
